Research on Polymorphism Relationship of Th17/Treg Cell and Its Factors with STAT4 Gene in AITD Patients with Different Iodine Nutritional Status

Abstract: Background: Research has shown that STAT gene SNP is related to the pathogenesis of T1D, SLE, RA and AITD. Objective: To investigate the correlation of the SNP of Th17/Treg cell and its cytokines with STAT4 in the PBMC of AITD patients with different iodine nutritious status. Method: 100 GD and HT patients who are selected from Cangzhou Central Hospital and Cangzhou People’s Hospital from September 2019 to August 2020, respectively, 60 healthy cases, adopt flow cytometer and quantitative real-time PCR to test the Th17and Treg cell ratio and proportion in PBMC and the expression level of ROR-γt and 3 (Foxp3) mRNA, and use ELISA method to test IL-17 and TGF-β level in the serum. Utilize SASP-PCR technology to test SNP locus (rs7574865) of STAT4 gene, and calculate the genotype and allele frequency. Result: There is some correlation between Th17/Treg cell & its factors in PBMC in the AITD patients and STAT4 rs7574865; compared with healthy control group, the difference between STAT4 rs7574865 G/T &T/T genotype and G&T allele frequency is of statistic significance (χ2 is 6.128 and 5.613 respectively, and the average value of P is <0.05), and may increase the occurrence risk of AITD, but has no correlation with iodine nutritious status. Conclusion: AITD patients not only have Th17 and Treg cell imbalance and the corresponding factor changes but also correlate to STAT4 rs7574865 susceptibility, and the cellular immunity and gene SNP may participate in the disease occurrence process of AIDT together.

Relationship Between the Expression of S100P in Gastric Cancer Tissue and Clinical Pathology

Abstract: Purpose: Discuss the diagnosis value and action mechanism of S100P for gastric cancer and the influence of S100P in tumor metastasis. Method: Collect 100 cases of gastric cancer tissues diagnosed as per histopathology by our hospital, apply immunohistochemistry method, test the positive expression condition of S100P protein, compare with 100 cases of control tissue specimens, and analyze the relationship between S100 protein expression condition in gastric cancer tissue and clinical pathology. Result: the positive rate (51.0%) of S100P in gastric cancer tissue is significantly lower than chronic atrophic gastritis with atypical hyperplasia, superficial gastritis and normal gastric mucosa (P<0.001 or P<0.05), while the difference between gastric cancer group and group of chronic atrophic gastritis with atypical hyperplasia has no statistic significance (P>0.05). The difference of positive rate of S100P in gastric cancer tissue with gender, age, tumor size, tumor position and Lauren typing has no statistic significance. In well differentiated adenocarcinoma, moderately differentiated adenocarcinoma, poorly differentiated adenocarcinoma and adenocarcinoma anaplastic, the positive rate (80.56%) of S100P of the three groups has significant difference (χ2=13.912, P=0.001). The positive rate of S100P which doesn’t invade serosa is significantly higher than the positive rate (34.38%, χ2=19.662, P<0.001) of S100P invading serosa and invading beyond the serosa, and the difference of positive rate of S100P of Phase I, Phase II, Phase III and Phase IV also has statistic difference (χ2=13.347, P=0.004), the positive rate (73.17%) of localized S100P protein is significantly higher than the positive rate (35.59%, χ2=13.669, P<0.001) of invasive type; the positive rate (74.42%) of S100P without lymph node metastasis is significantly higher than the positive rate (33.33%, χ2=15.020, P<0.001) with lymph node metastasis. Conclusion: the positive rate of S100P protein in gastric cancer tissue is significantly lower than the para-carcinoma tissue; the lower the differentiation degree is, the deeper the invasion is; the patients who have later TNM staging and Borrmanntyping, and lymph node metastasis have lower positive rate of S100P protein, which indicates that the low expression of S100P protein participates in the occurrence, development, invasion and metastasis of gastric cancer, and is an independent dangerous factor affecting the patients’ prognosis.

Abstract: Background: Medical students throughout the world show a high rate of sharp injuries with a consequent risk of acquiring blood-borne infections while performing their clinical activities. Objective: This study was conducted to assess needle stick or sharp injuries among medical students of Debre Tabor University, and to identify the associated factors leading to Needlestick or sharp injury. Method: An institution-based cross-sectional study was conducted among 4th to 6th-year medical students at Debre Tabor University, by using census sampling through a self-administered semi-structured questionnaire, and the completeness of the questionnaires was assessed daily. Data were processed and analyzed using SPSS software version 21.0 and bivariate analysis was obtained using a logistic regression model. In addition, tables and figures were used as necessary to represent the data. Result and conclusion: According to our study, the prevalence of Needle stick or sharp injuries among medical students in clinical years other than clerkship I students in Debre Tabor University was found to be 28.5%. Medical interns were particularly affected with a prevalence rate of 66.6%. The associated factors with Needlestick or sharp injuries included the academic year of study and the department where the injury occurred.

Abstract: Background and objectives: Coronavirus (COVID-19) is pandemic, has affected almost every country around the world. A retrospective multi-centered study was carried out to determine the frequency of COVID-19 in children and adults in Lahore, Pakistan from March 2020 to May 15 2020. The study was included 574 corona positive (PCR–positive) patients of all age groups including 147 children. Record of the patients who visited emergency with symptoms of Corona like fever, sore throat, and respiratory distress and those admitted in hospitals due to corona illness was analyzed. Travelling history was recorded and frequency determined using SPSS ver 21. Data of study showed that 54% middle aged patients were found to suffer with Corona virus followed by children and 23% old age group. The patients belong to middle / low socioeconomic status. 26 patients had international travel history while 49 travelled domestically. Study concluded that the incidence of coronavirus disease was more in middle age men followed by children and old age group. However, it is found that the incidence of virus observed in children with better recovery than other age groups. In most of the cases, the course of disease is mild to moderate with less mortality rate.
